First, read the Documentation. There is a lot of useful stuff in there and a lot of your initial questions are bound to be answered there.
This page was started as a kind of guide for those of a less technical bent who would like to get to grips with MediaWiki. Any aspects of running a wiki are fair game just as long as you use non-technical language (the less jargon the better). If you've found a page which helped you get your wiki up and running post it here.
